MRC Holland and Amsterdam UMC have launched a collaborative project, supported by Health~Holland, and aimed at improving the reliability of patient-derived stem cell models used in personalized medicine. These models are increasingly important for studying diseases such as neuropsychiatric disorders, but genetic changes acquired during cell culture can compromise their accuracy and limit their value for research and future therapeutic applications.
The project aims to utilize the high throughput of our NGS-based digitalMLPA technology to create an affordable and scalable solution that will enable researchers to routinely assess the genomic stability of their cells while in culture and during length differentiation protocols.
By combining Amsterdam UMC's expertise in stem cell biology with MRC Holland's innovative genomic technologies, the collaboration aims to deliver a validated workflow, including protocols, quality control guidelines, and recommendations for broader implementation in both academic and industrial settings. The project will form the basis of a PhD project, jointly supervised by MRC Holland and the research group of prof. Vivi Heine from Amsterdam UMC.

Preliminary evidence presented at ISSCR 2026 in Montreal (Canada) from tests performed at MRC Holland with our NXtec D024 KaryoProfiler assay confirms that digitalMLPA could indeed be a promising tool for high-throughput cell line quality control. The assay was able to detect aberrations in cell culture samples at a 2-4 Mb resolution, producing consistent results with aCGH and Karyostat data. A valuable feature of the assay is the 39-digit SNP code which provides a unique profile that aids in cell line authentication and cross-contamination checks.
The data, which was presented during a poster session and in a corporate talk in the exhibition hall, sparked a lot of interest and led to many interesting discussions that confirmed to us that we are on the right track. So stay tuned for further developments on this topic!
