Documentation Update - digitalMLPA General Protocol

We’ve recently updated the digitalMLPA General Protocol to reflect a new recommendation for read depth. For optimal results, we now recommend a median read count of at least 600 for all reference probes in each reaction, up from the previous threshold of 500. The absolute minimum read depth remains at 400, but we advise using 600 to avoid potential warnings related to median total reads and read depth during analysis with Coffalyser digitalMLPA. 

Important notes:

This change in the protocol will have a slight effect on the maximum number of samples that can be combined in a single sequencing run. The maximum sample count depends on several factors:

  • The number of probes in the probemix(es) used
  • The capacity of the Illumina reagent kit
  • The number of barcode solutions available
