Modifications to reagents in Ligase-65 kits

From March 2019 onwards, the reagents in all Ligase-65 kits will change. There will be a small change in the storage buffer of the SALSA Ligase-65 enzyme. The SALSA MLPA Buffer, which is included in the kit with product code Lig-5a, will also be modified.

What will change?

The β-mercaptoethanol antioxidant in the storage buffer of the Ligase-65 enzyme will be replaced by the antioxidant dithiothreitol (DTT) because this is less pungent and less toxic. The enzyme itself will not change.

The new MLPA buffer also contains DTT. This counters sloping caused by iron ions in the sample DNA, which is a common impurity in DNA samples extracted with magnetic bead-based systems. The MLPA buffer is only included in the kit with product code Lig-5a.

We do not expect these changes to affect applications other than MLPA. However, you may need to revalidate your assay.

Timeline

Starting March 1st 2019, all Ligase-65 kits will contain the new reagents. Old reagents are available on request between March 1st and August 31st 2019. Use reference SMR42 to order the old MLPA buffer, and SMR48 to order the old Ligase-65 solution.
